In France, marine fish and shellfish farming is based mainly on the breeding of wild species, whose natural populations are traditionally exploited. Research programs are conducted by IFREMER, in co-operation with INRA and Universities. French research programs for the genetical improvement of marine fish and shellfish concern primarily seabass (Dicentrarchus labrax) and oysters (Ostrea edulis and Crassostrea gigas). Haplosporidiosis in the pacific oyster Crassostrea gigas from the French Atlantic coast ArchiMer

Two cases of haplosporidian infection occurred during 1993 in Pacific oysters Crassostrea gigas from the French Atlantic coast. The localization and ultrastructure of the plasmodia are described. In situ hybridization of infected tissue sections was conducted with DNA probes for oyster-infecting haplosporidians. The Haplosporidium nelsoni-specific DNA probe MSX1347 hybridized with the C. gigas parasite, and the H, costale-specific probe SSO1318 did not hybridize. Total genomic DNA was extracted from the infected tissue sections for polymerase chain reaction (PCR) amplification of the haplosporidian. L'aquaculture et la pêche ne font pas toujours bon ménage : conflit pour les zones de production (élevages en mer), concurrence sur le marché (poisson d'élevage / poisson "sauvage"), et surtout conflit sur la gestion des stocks naturels de juvéniles, parfois exploités pour l'approvisionnement des élevages au détriment d'une production naturelle d'adultes pour le stock exploité par pêche (palourde du golfe du Morbihan, crevettes tropicales, etc). The Aquaculture and Pathological Genetics (GAP) laboratory at La Tremblade has been monitoring three populations of European flat oyster since 1985 or 1989 under selection for higher survival rates in the presence of the parasite Bonamia ostrea. One selection cycle (1985 population) or two (1989 populations) were carried out by mass selection by placing the oysters for several years in a highly infested zone, and then harvesting spawns from survivors.

Image analysis : a new method for estimating triploidy in commercial bivalves ArchiMer

For determination of ploidy levels in bivalves, three techniques are now routinely used: karyological determination. microfluorimetry and flow cytometry. This paper proposes an alternative technique which is based on determination of the optical density (OD) of stained nuclei using image analysis. A karyological and image analysis comparative experiment conducted on juveniles of Crassostrea gigas (Thunberg) and Ruditapes philippinarum (Adam & Reeve) is described and highlights the advantages of the image analysis technique. In order to study individual growth variability and its relationship with survival in juvenile Crassostrea gigas, parental oysters were sampled at four sites located along the French Atlantic coast and bred under controlled hatchery conditions. Four groups of larvae were obtained by crossing five males and five females from each of the four sites, and a fifth group by crossing these 20 males and 20 females together in a pool. Induced triploidy in the European clam, Ruditapes decussatus (L.) and performance of triploid larvae ArchiMer

For the first time, effective treatments using cytochalasin B were developed to induce triploidy in the European clam, Ruditapes decussatus (L.). The percentage of triploid embrios was assessed by karyological or image analysis. Two treatments (O.5 or 1 mg of cytochasin B (CB per ml of dimethylsulfoxide in 1 l of sea water) were applied at different times after fertilization (10 to 25 mn), for two different periods (15 and 20 min). Best results were obtained for a CB concentration of 1 mg/l. When treatment was applied 15 min after fertilization for a 20 min period, 94% and 95% of triploid embryos were obtained in two repeated experiments. Influence of parentage upon growth in Ostrea edulis: evidence for inbreeding depression ArchiMer

Genetic variability for growth was analysed in three populations of Ostrea edulis, selected for resistance to the protozoan parasite Bonamia ostreae. This study was undertaken first to determine the potential for selection for growth in populations that have never been selected for this character, and second to estimate heterosis versus inbreeding depression. The present study was designed to compare the response to contaminants in diploid with triploid specimens of the oyster Crassostrea gigas. The reproduction investment in bivalve molluscs has priority on somatic growth. Thus, genetic sterilization by triploidy induction enables the energy flux to be directed toward somatic growth and glucide storage. The European flat oyster Ostrea edulis has been considered as the traditional French oyster for a very long time and has now nearly disappeared from our farms, as a consequence of the two successive epizootic of the Marteilia refringens parasite and Bonamia ostreae. The actual quasi monoculture of Crassostrea gigas has made necessary the search for a diversification in oyster culture, including the search for a new start of the indigenous flat oyster culture. The Ifremer has been conducting parallel works since 1985 in order to better understand the biology of the parasite as well as to try and select different lines of flat oyster resistant to Bonamia ostreae.

The king scallop (Pecten maximus L.) has been the object of some in-depth studies since 1973 with the double aim of natural resources management and extensive aquaculture in mind. The results obtained during the first ten years enabled the launch of a pluri-annual 1983-1987 program which aims to demonstrate the re-populating of the roadsted of Brest and the introduction of new management methods in the Bay of Saint-Brieuc. The technical results of the resource management and of the aquaculture operations are described. They enabled the stabilisation of the annual production of the Saint-Brieuc deposit as well as to foresee a new biomass of 80-100 tonnes of spawners per year in the roadsted of Brest. Optimization of triploid induction by the use of 6-DMAP for the oyster Crassostrea gigas (Thunberg) ArchiMer

A novel method using 6-dimcthylaminopurinc (6-0MAP) has recently been demonstrated to be a good triploid inductor for bivalve molluscs. The effects of 6-DMAP concentration, timing of treatment after fertilization and treatment duration on survival and triploid induction of Crassostrea gigas (Thunberg) embryos were examined. An influence of sperm concentration was also detected and discussed. Survival to D-stage was inversely related to 6-DMAP concentration and the percentage of triploids was shown to be 6-DMAP dose dependent. Two optimal moments for treatment application were determined corresponding to extrusions of the first and second polar bodies.
